Open Lab of Advance Microscopy - OLMA@MBC

Providing training and assistance for state-of-the-art microscopy

OLMA@MBC is an open access facility of the Molecular Biotechnology Center (MBC) of the University of Turin. The facility allows access to state-of-the-art equipment including widefield and confocal microscopes for the structural and molecular analysis of live and fixed cells or tissues.

OLMA@MBC provides support and user training on all equipment. In addition to microscopy equipment, we also provide access to software and expertise in image data analysis and general microscopy education

We provide assistance throughout the process of microscopic imaging:

  • Training of new users.
  • Technical assistance.
  • Support for image processing, visualization and analysis.
  • Support for advanced microscopy techniques.

The facility is open to scientific collaborations on specific projects.

Equipment

Leica STELLARIS 8 inverted confocal microscope

  • Lasers: 405 nm, tuneable White Light Laser (440-790nm) for imaging wide-range of fluorophores from blue to near infrared.
  • 3 high-sensitivity and low-noise spectral HyD detectors, detection range up to 850nm.
  • Conventional and high speed resonant (8000 Hz) scanners.
  • Objectives: 10x/ 0.3, 20x / 0.75 dry, 40x / 1.3 oil, 63x / 1.4 oil.
  • Incubation system for live cells.
  • Motorized xy-stage for imaging multiple areas per dish.
  • Auto Focus Control.
  • TauSense technology allow to take advantage of average photon arrival times.
  • Lightning Confocal Super Resolution Modality
  • LAS X software with 'LAS X 3D' image viewer and 'LAS X Navigator' for convenient tile-scanning and stitching

Leica SP8 inverted confocal microscope

  • 8 excitation laser lines: 405, 458, 477, 488, 496, 514, 561, 633 nm.
  • 2 PMTs fluorescence detectors and 2 sensitive HyD detectors for increased sensitivity and photon-counting experiments.
  • AOBS based system.
  • Objectives: 5x, 20x / 0.75 dry, 40x / 1.3 oil, 63x / 1,4 oil.
  • Incubation system for live cells.
  • Motorized xy-stage for imaging multiple areas per dish.
  • Auto Focus Control.
  • LAS X software with 'LAS X 3D' image viewer and 'LAS X Navigator' for convenient tile-scanning and stitching

Leica SP5 inverted confocal microscope

  • 7 excitation laser lines (405, 458, 477, 488, 514, 561, 633).
  • Conventional and high speed resonant (8000 Hz) scanners.
  • 3 PMTs fluorescence detectors and 2 sensitive HyD detectors for increased sensitivity and photon-counting experiments
  • Objectives: 20x / 0.5 dry, 20x / 0.75 imm., 40x / 1.25 oil, 63x / 1.4 oil.
  • Incubation system for live cells.
  • Motorized xy-stage for imaging multiple areas per dish.
  • FRAP/FLIP/Photoactivation, FRET
  • Leica LAS AF matrix for multiple acquisitions/ multicontent imaging.

Fluorescence stereomicroscope Leica MZ 10 F

  • Brightfield/darkfield transmitted/reflected light.
  • Red, green and blue fluorescence.
  • 8x to 80x total magnification.

Leica DM6 B 

  • Automated upright microscope.
  • Transmitted light images – brightfield.
  • Motorized xy-stage for tiling/stitching.
  • Colour camera for IHC, H&E, etc. staining.
  • LAS X software with 'LAS X Navigator' for convenient tile-scanning and stitching.

THUNDER Imager Live Cell

  • Inverted Leica DMi8 fully motorized with digital clearing.
  • Motorized x-y stage
  • Epifluorecence illuminator with 8 single LEDs (395, 438, 475, 511, 555, 575, 635, 730nm)
  • Leica K8 sCMOS monochrome camera.
  • Objectives: HC Plan Fluotar 4x/0.13 PH0, HC Plan Apo 20x/0.80 PH2, HC Plan Fluotar 40x/0.60 CORR PH2, HC Plan Fluotar 63x/0.10 IMM CORR PH3
  • Leica Thunder software module includes Instant Computation Clearing (ICC) Small Volume Computation Clearing (SVCC) and Large Volume Computational Clearing (LVCC)

Zeiss Axio Observer

  • Inverted microscope.
  • Phase contrast.
  • Filter sets for a wide variety of fluorophores.
  • 5x dry to 100x oil objectives.
  • Slide and multi-well plate holder.

Image analysis

Facility users have access to a powerful workstation dedicated for image analysis and visualization. Several software packages are installed to support a variety of data formats.   These tools support image processing, intensity measurements, object counting, and the creation of movies and 3D models.

  • Z2 Tower G9 Workstation.
  • Intel Core i7 -14700 20 cores.
  • 128GB of RAM.
  • Nvidia RTX A4000 16 GB graphic card.
  • SSD 8TB hard drive.
  • Fiji (ImageJ): an open-source software for microscope image analysis. With Fiji, you can visualize your results, utilize custom plugins, and create your own analysis macros for automated image processing and quantification.
  • Imaris (Bitplane / Andor): powerful 3D and 4D image analysis software, enables scientists to explore complex biological data, visualize cellular structures, track dynamic processes, and quantify features within multidimensional microscopy images. We offer two floating licenses.
  • Leica LAS X off-line version: you can view and process your images acquired with Leica confocal microscopes.
